I don't know what has Oda said about this (couldn't find anything) so... here's a theory- what if the will of D. is connected to the great kingdom against whom 20 others united to fight. D in my opinion can symbolize the name of the kingdom or the ruling family of that kingdom as it is known that they are enemies of celestial dragons. And they are found in all the seas so they probably escaped to all parts to survive after the war. Maybe joyboy was the ruler of that kingdom and the last island is that only, it's his home. World government tried to erase it off the map too and they could easily have lied that it was destroyed completely but a part of it remains. Joyboy is known to spread happiness and carriers of D. die with a smile on their face so it's highly likely that all 3 of these are connected.

Guys, i dont know much but what if one piece is not material wealth, maybe there is money there but no one can access it, and the real treasure is all these experiences and possibly the information present on that island. And the reason the island is called laugh tale is because joyboy knew how hard it was to get there and left money there but made it inaccessible to anyone and wrote-
"The strength, friends, knowledge etc. you gained while trying to get here is the one piece, this money is worthless. Also- here is the information about the world which is being erased by the winning group, I wish they never find this so that true history is never erased completely."
And using some powers or whatever he made that island get isolated from magnetic fields and all so that compasses of all kinds are useless, and he made the island practically invisible. You absolutely have to do some things (like collecting poneglyphs) so that you actually have that experience.
And this is what made Roger laugh, that all he did was the real one piece, it was never about the island and material wealth in the first place!
(Shanks is now making his move to claim one piece, so maybe that wealth is accessible using awakened hito hito no mi model: nika's powers, he may know that)
